- Label flask with cell line, passage number, date
- Put 10 mL medium in flask, set in incubator
- Retrieve frozen vial from LN2 dewar, thaw in incubator or water bath.
- Put ~1 mL thawed cells into a single flask
- 24 hours later, replace medium (5 mL)
- Split as needed
